The dead man, named locally as 26-year-old Sandy Kelly, was discovered in Bute Drive at the junction with Gowans Terrace shortly after 18:30.
He was taken to Perth Royal Infirmary where he died. It is understood he had a single stab wound to the chest.
Tayside Police said the arrested man was detained in the centre of Perth at about 15:30.
The death is being treated as murder, but a force spokesman said inquiries into the circumstances surrounding the incident were continuing.
The attack happened directly outside the grounds of Perth Grammar School.
Officers have been making door-to-door inquiries and reviewing public and private CCTV coverage of the area where the man was found.
The force appealed to anyone with CCTV installed who lived or worked near the scene of the attack to contact them.
Supt Tony Beveridge said: ''We want to speak to anyone who was in the area at or around 6.30pm last night as they could have information useful to our enquiries.
''There were a number of people in the area last night, as well as several vehicles that passed through where the incident took place. We want to speak to those people if they haven't spoken to us already."
Supt Beveridge, deputy divisional commander for Perth and Kinross, added: "Uniform police officers will be on high visibility patrols in the area to offer reassurance to the community.
"Similarly, anyone who has information that could help us should approach our officers and speak with them.''
