Firefighters were called to the blaze in Riverford Gardens, Pollokshaws, just after 19:00 on Friday.
At the height of the incident, 15 fire engines were at the scene. It is believed the building was a block of flats in the process of being built.
Police are appealing for information about three youths seen in the area at about 18:00 on Friday.
Det Con Alyson Dowds said: "We are working to identify the three people who were seen entering Riverford Gardens around 6pm on Friday evening, they were in the area for about 15 minutes then left heading towards Riverford Road."
She said they were wearing dark hooded jackets. One of them had fair hair and one had dark hair. Two of the youths were wearing dark trousers with the other one wearing light-coloured jogging bottoms.
"We have been carrying out inquiries in the local area and studying CCTV footage to gather more information," Det Con Dowds added.
She urged anyone who may have seen the youths to contact police.
In the early hours of Friday a man died following a flat fire in Parnie Street in the Trongate area of the city.
Police said the man was brought out of the building and treated by ambulance staff but was pronounced dead at the scene.
Investigations are under way to establish the cause of the fire.
