Leicestershire's Matthew Boyce made a career-best century but Gloucestershire hit back in the final session on day three at Grace Road.
Boyce (119) made three figures for only the second time in 52 first-class games, putting on 195 with Will Jefferson (90) for the first wicket.
But the hosts, 200-2 at tea, were unable to capitalise and closed the day on 315-9, still 189 runs behind.
Ian Saxelby (3-66) and Jon Lewis (3-47) led the fightback for the visitors.
