Will Smith's third half-century of the season has given Durham a solid platform in reply to Somerset's first-innings total of 480 all out.
The visitors closed on 150-2, trailing by 330, with Smith unbeaten on 58.
Earlier, Somerset added 71 to their overnight score to eventually be bowled out for 480.
Murali Kartik (43) and Alfonso Thomas (42) took their eighth-wicket partnership to 83, while Mitch Claydon claimed career-best figures of 6-104.
Graham Onions and Ian Blackwell both finished with two wickets each, while Steve Harmison, who took 0-65 in 10 overs on day one, was not used on the second morning.
Durham's reply got off to a solid start with Smith and Michael di Venuto (46) putting on 84 for the first wicket.
Steve Kirby (2-30) had the latter caught by Arul Suppiah and soon accounted for Gordon Muchall for eight.
Paul Collingwood (28 not out) and Smith then put on an unbroken third-wicket stand of 58 to see Durham to stumps on 150-2.
