Glamorgan will go into the final day still needing 119 runs to make Division Two leaders Northants bat again as they battle to save the game at Swansea.
After starting the day on 434-4, Northants eventually declared on 552-9, Stephen Peters extending his overnight first-ever double century to 222.
But Glamorgan, bowled out for 252 first time round, dug in to make 181-3 by the close, led by Alviro Petersen (75),
The Glamorgan skipper put on 136 for the second wicket with Will Bragg (69).
Northants' total was the biggest by any county side at St Helen's, beating Leicestershire's 536 in 1996, making them still firm favourites to secure a sixth win of the season.
Peters' 492 minutes in the middle for a career-best 222 from 390 balls earned him the seventh-highest individual score on the ground.
