Martin Guptil hit 86 as Derbyshire built a lead of 112 after Essex were 207 all out in their first innings.
Essex's Ryan ten Doeschate added just one to his overnight score, falling on 55 before Maurice Chambers and Reece Topley added a further 33 runs.
Derbyshire lost debutant Matt Lineker for 16, but Wes Durston (62 not out) joined Guptil and the pair added 95.
The opener was bowled by ten Doeschate while Masters dismissed Dan Redfern to leave the visitors on 187-4.
