Kane Williamson, Richard Coughtrie and Chris Dent all scored half-centuries as Gloucestershire compiled a solid 278-2 in response to Glamorgan's 378 all out.
Earlier, Graham Wagg was left stranded on 70 as the visitors were bowled out, with David Payne finishing on 4-79.
Dent and Coughtrie put on a century partnership before Coughtrie was dismissed for 50 by Jim Allenby.
Dent was run out by Wagg on 81 - Williamson and Chris Taylor were 84 and 42 not out respectively at stumps.
