Jimmy Adams and Michael Carberry shared a 373-run partnership, a new second-wicket record for Hampshire, on their way to posting 480-4, a lead of 276.
Resuming on 124-1, the pair batted serenely for the top flight's bottom club before Carberry went for 182.
His dismissal was followed by two more quick wickets before Sean Ervine arrived to partner Adams.
The Hampshire captain finished the day on 204 not out, with Ervine unbeaten on 56, Steve Kirby picking up two wickets.
