Derbyshire spinner David Wainwright claimed his third five-wicket haul in six Championship matches to help his side take control at Chelmsford.
Former Essex skipper Mark Pettini top scored with 56 as the home side were bowled out for 182.
It was the 28-year-old right-hander's fourth half-century in successive Championship games.
Derbyshire responded by then reaching 37-1 to close a day dominated by former Yorkshire left-arm spinner Wainwright.
Varying his flight intelligently, Wainwright, let go from Headingley at the end of last season, claimed a wicket with the first ball of all his three spells.
And he also got rid of Pettini, hanging on to a fierce drive for a return catch that completed his latest five-for.
