Northants made 531 then reduced Essex to 187-5 at the close on day two.
Resuming on 343-5, thanks to centuries from David Sales and Stephen Peters, Andrew Hall and Steven Crook scored 63 apiece as the visitors went beyond 500.
Essex then lost Jaik Mickleburgh early and after recovering to 65-1, collapsed to 103-5 - Crook dismissing debutant Gautam Gambhir and Ravi Bopara cheaply.
But Ryan ten Doeschate (47no) and James Foster (39no) rallied, shepherding the hosts to stumps.
