England captain Alastair Cook made 181 to put Essex in a strong position going into day four against Derbyshire.
Cook kept ticking over before being caught and bowled by David Wainwright, and soon after Essex were out for 425, setting the visitors a target of 336.
Derbyshire openers Stephen Moore (32) and Billy Godleman (38) gave their side a relatively solid footing, but Tymal Mills (3-29) began to show his prowess.
The visitors closed on 167-5, but may now rely on Shiv Chanderpaul (41 no).
Chanderpaul was the only batsman left standing in Derbyshire's first innings, which saw the last six wickets fall for just 19 runs - three of those going for nought.
