Leicestershire's Nathan Buck took four wickets to put Hampshire 260-7, but the visitors fought back to close on 321-8.
Hampshire lost wickets regularly after an opening stand of 75, James Vince (70) and Adam Wheater (57) providing the only other noteworthy partnership.
Buck snared two in quick succession to make it 219-6, and Sean Ervince (40) became his fourth scalp after rallying.
Kyle Abbott (33 not out) and Danny Briggs (14 not out) then frustrated the hosts, surviving 45 minutes to stumps.
