Northants all-rounder David Willey took three wickets in an over as they bowled out Gloucestershire for 296 on day two at Wantage Road.
Willey removed James Fuller, Craig Miles and Liam Norwell in the space of four balls as he claimed 4-72.
Geraint Jones was bowled for 80 by Rory Kleinveldt in the next over as Gloucestershire lost their last four wickets for no runs in seven balls.
Already ahead by 37 on first innings, Northants were 68-2 at close of play.
Pace bowler Craig Miles followed his 6-63 in the home side's first innings by having Richard Levi (13) caught behind and bowling Stephen Peters (16) in successive overs.
And the visitors will look to him to make early inroads on day three, when Northants skipper Alex Wakely will resume on 21 not out, with partner Rob Newton unbeaten on 10.
Gloucestershire captain Geraint Jones:
"Although we're disappointed to lose four wickets at the end in quick succession, make no mistake, Willey's bowling at the death was top-class.
"There's still something in the pitch and if we can get some early wickets tomorrow morning, we've got a chance to still win this.
"I was pleased by my own batting performance."
Northants all-rounder David Willey:
"I thought it might swing and pitched it up. I knew we were always going to be in the game with the way that Jones and Fuller batted, both going for their shots.
"I was lucky to hit the right area at the right time.
"The first hour tomorrow is going to be massive. If we can get through that, we will be a in a really strong position."
