Ashwell Prince made a timely century as Lancashire responded well to Northants' tail-end rally in the morning session.
Resuming on 198-6 after a rain-hit first day, Northants went on to pile up 385, sparked chiefly by Steve Crook, who made 91 against his former county.
He shared an 80-run seventh-wicket stand with Adam Rossington (57), further aided by Olly Stone (38).
Lancashire then reached 216-4 by the close, with Prince unbeaten on 104, his second hundred in successive innings.
It was his 42nd first-class century, having hit 106 in Lancashire's nine-wicket win over Kent last week.
Northants all-rounder Steven Crook:
"It's a good wicket to bowl on. I think it may be a bit up and down over the next couple of days, but you've got to be right on the money with the ball or you end up round the park.
"We've got to go in on Tuesday morning and, with a couple of wickets for us, we're right in the game."
Lancashire batsman Ashwell Prince:
"I thought Horton played well again today and that partnership was obviously crucial after the two early wickets.
"Then Alex Davies showed his talent too. He's a gutsy player, a good young player and he was aggressive in playing his strokes."
