Thirteen wickets fell between Durham and Hampshire on day one at Chester-le-Street as bad light ended play early.
The hosts were reduced to 65-5 before Ryan Pringle shared a 69-run sixth-wicket stand with Gordon Muchall (23).
But, the 23-year-old fell one run short of his maiden ton as Durham were bowled out for 213, as Ryan McLaren took 4-60 and spinner Gareth Berg claimed 3-27.
In reply, Hampshire closed 136 behind on 77-3 with Graham Onions taking 3-32, including James Vince for just nine.
Having announced they will sign Essex and England bowler Reece Topley at the end of the season, Hampshire handed debuts to McLaren and Ryan Stevenson.
Under sunny conditions Durham's decision to bat first turned out to be a poor one with West Indian paceman Fidel Edwards also claiming 3-57, as wicketkeeper Adam Wheater took four catches behind the stumps.
Pringle hit 19 fours in his mature innings on his way to a career-best first-class score, but afternoon cloud cover ensure the bowlers remained on top.
The visitors currently sit bottom of Division One with four matches left to play and need to win to keep in touch with seventh-placed Sussex.
However, Durham have little left to play for this season having been ruled out of the title race and looking unlikely to be dragged into a relegation battle.
