Terry Gornell's 90th-minute goal saw off Oxford and ended Accrington's five-game League Two losing streak.
Adam Buxton hit the bar from distance before Gerardo Bruna rifled a shot against the post as the hosts searched for a goal their play deserved.
Oxford offered little in attack but James Roberts missed their best chance, scuffing over from five yards out.
The winner came when Gornell hooked the ball over his shoulder after Kal Naismith's cross was not cleared.
