Mike Jones scored the only goal of the game with a stunning free-kick from 25 yards after Jonathan Forte was pushed by Chris Robertson.
Vale looked to respond, but Jake Kean denied Ben Williamson before Tom Pope saw his effort cleared off the line.
Oldham held on to secure the three points and end Vale's three-match winning sequence to rise to 11th.
