Jai Reason put the Spitfires ahead shortly before the break, turning his marker and firing past Andy Coughlin.
Southport were reduced to 10 men after Andrai Jones' two-footed lunge on Joe Partington after 49 minutes.
Eastleigh capitalised on the extra-man advantage as Jamie Turley, Ben Strevens and James Constable all added to the goal tally in the second half.
