Jamie Proctor's first-half effort was enough to earn Fleetwood all three points away at struggling Barnsley.
The striker, 23, collected the ball inside the opposition half, before out-foxing Tykes defender Marc Roberts and slotting past Nick Townsend.
Barnsley came close to equalising when Fleetwood keeper Chris Maxwell was forced to make a fingertip save to keep out Alfie Mawson's free-kick.
Lloyd Isgrove then had an effort blocked as Fleetwood just held on.
Victory for the visitors saw them leapfrog their opponents and rise out of the relegation places, with Barnsley slipping into the bottom four.
