Clermont Auvergne lock Jamie Cudmore and Ospreys pair Tyler Ardron and Jeff Hassler feature in Canada's line-up to face Scotland on Saturday.
Cudmore worked with Scotland head coach Vern Cotter when the latter was with Clermont between 2006 and earlier this year.
London Scottish's Tyler Hotson and London Irish's Jebb Sinclair also feature in Kieran Crowley's side.
Glasgow Warriors back DTH van der Merwe is listed among the Canada substitutes.
Canada will meet Scotland's fellow Six Nations sides France, Ireland and Italy as well as Romania in next year's World Cup in England.
But flanker Sinclair insists Crowley's players are not looking that far ahead.
"None of us are thinking about next year's World Cup; all our attention is focused on Saturday and we're hoping to replicate our fast start from against Japan," Sinclair told the Rugby Canada website.
"We need to fix a few defensive errors that let us down in the second half.
"I think Scotland's game plan matches up well with how Canadians like to play - big, direct, hard ball runners - and they have a strong set piece that they've turned around recently under Cotter.
"We're excited for the challenge and looking forward to playing in front of a packed house at BMO Field on Saturday."
